描述.Originally confused with opioid receptors and then orphan receptors with no biological function, Sigma Receptors are now recognized as relevant to many degenerative diseases with remarkable potential as therapeutic targets. In this text, new information about the structure of sigma 1 receptor, its binding sites are provided as well as its expression in many cell types.  It’s putative role in degenerative neuronal diseases including amyotrophic lateral sclerosis, Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, pain, drug addiction and locomotor activity. Their roles in possible treatments for blinding retinal diseases emphasize the tremendous far-reaching potential for ligands for these receptors.  Exciting breakthroughs in this dynamic field in the last decade are reported herein, which will guide future investigators in determining the full potential of this unique, yet abundantly expressed protein..

,The Evolution of the Sigma-2 (σ,) Receptor from Obscure Binding Site to , Therapeutic Target,o binding studies over 25 years ago, the molecular identity of this protein is currently not unambiguously known, and the results from recent attempts to identify the σ. receptor through protein purification and mass spectral analysis have been the subject of debate in the literature. However, there

Pharmacological Modulation of the Sigma 1 Receptor and the Treatment of Pain, ies and reduce their unwanted effects. Current preclinical evidence supports the modulatory role of sigma-1 receptors (σ.R) in nociception, mainly based on the pain-attenuated phenotype of σ.R knockout mice and on the antinociceptive effect exerted by σ.R antagonists on pains of different etiologies

Sigma-1 Receptors and Neurodegenerative Diseases: Towards a Hypothesis of Sigma-1 Receptors as Ampldiseases. Accumulating evidence indicates that sigma-1 ligands can either directly or indirectly modulate multiple neurodegenerative processes, including excitotoxicity, calcium dysregulation, mitochondrial and endoplasmic reticulum dysfunction, inflammation, and astrogliosis. In addition, sigma-1 l
