描述The advent of hybridoma technology leading to the successful produc tion of monoclonal antibodies against a variety of tumor-associated antigens has, during the last decade, provided a very powerful tool for research and clinical investigations. These highly specific reagents have essentially replaced the polysera of the earlier days. The successful demonstration of the many wide ranging capabilities of the monoclonal antibody technique has already begun to exert an enormous impact on diverse areas of research in basic science and medicine. In particular, the potential of monoclonal antibodies to serve as carriers for selective targeting of radionuclides to tumors for diagnosis or therapy, has stimulated an intense surge of research interest and even revived hopes of realizing Ehrlich‘s concept of the "magic bullet". Indeed, the technology appears to be on the threshold of a revolution in diagnosing and treating malignant disease. Much work remains to be done, however, and even though the progress has been impressive, results to date have shown only moderate success. There is no question that the limited success we have achieved thus far is merely a prelude to the many more exciti

Immunochemistry of Hybridomaseasibility of manipulation for specific purposes. Their use, however, is limited because of structural and physicochemical idiosyncrasies, occasionally restrictive factors influencing affinity and avidity, unique cross-reactivities, lack of multivalency compared to polyclonal antibodies, and restric

Characterization and Selection Criteria of Monoclonal Antibodies for Tumor Imaging Studiese the result of alterations in the gene regulation mechanism leading to either a blockage, stimulation or neosynthesis of cell surface molecules which, under normal conditions, would mediate cell-cell interaction and cellular differentiation (1,2). Thus, cells which have been oncologically transform

Selection and Utilization of Monoclonal Antibody Defined Melanoma Associated Antigens for Immunoscinrmation of melanocytes (for review, see 1,2). These antigens are referred to as melanoma associated antigens (MAA). Most of them, if not all of them, are detectable in melanocytes which are cultured . and are likely to be differentiation antigens.

Development and Characterization of Human Monoclonal Antibodies and Their Application in the Radioimse. Antibodies labeled with radioactive isotopes have been demonstrated to localize in tumors of the gastrointestinal tract (1–6), ovary (6), breast (6,7), and skin (8) and promise to be very helpful in identifying metastases in patients with these tumors. However, there remain several problems to b

Progress in Diagnostic Immunoscintigraphy and First Approach to Radioimmunotherapy of Colon Carcinomes injected into a tumor bearing animal can localize preferentially into the tumor, which could be detected by external scanning. These early studies, however, were performed with poorly characterized antibodies obtained by absorption and elution from tumor cells or crude tumor fractions. Thus, this

Immunohistochemistry and Correlation with Plasma Levels of CA 19-9 and CEA in Gastrointestinal Tumorn and follow-up of malignant tumors has grown steadily (1). The antigenic determinant most likely resides in the peptide portion of the molecule. The specificity of CEA however, is influenced and diminished by various other glycoprotein antigens cross reacting with antisera to CEA. Utilizing hybrido
