描述.Over the last decade it has been shown that orphan G protein-coupled receptors (GPCRs) can be used as targets to discover novel neuropeptides. A dozen neuropeptides have been identified through this approach. Each of these neuropeptides has opened new doors for our understanding of fundamental physiological or behavioral responses. For example the orexins, MCH and ghrelin carry fundamental roles in regulating food intake while neuropeptide S, neuromedin S, the prokineticins and the orexins are major players in modulating sleep and circadian rhythms. ...The chapters of this book review the latest research in the field, most of them are written by the original discoverers of the respective novel neuropeptide. Emphasis is set not only on their discovery but also on their functional significance. Since many of these neuropeptides are part of drug discovery programs, this book impacts academic as well as pharmaceutical research..

Orexins and Orexin Receptors: From Molecules to Integrative Physiology,y results in narcolepsy-cataplexy in humans, dogs, and rodents, suggesting that the orexin system is particularly important for maintenance of wakefulness.Orexin agonists and antagonists are thought to be promising avenues toward the treatment of sleep disorders, eating disorders, and drug addiction.

Prolactin-Releasing Peptide, ase from cells stably expressing the orphan G protein-coupledreceptor hGR3 (Hinuma et al. 1998) [also known as GPR10 (Marchese et al. 1995), or UHR-1 forthe rat orthologue (Welch et al. 1995)]. Initially touted as a prolactin-releasing factor (thereforeaptly named prolactin-releasing peptide), the p

Structure and Function of Ghrelin,e”) in Proto-Indo-Europeanlanguages meaning “grow”, since ghrelin has potent growth hormone (GH) releasing activity. Inaddition, ghrelin stimulates appetite by acting on the hypothalamic arcuate nucleus, a region knownto control food intake. Ghrelin is orexigenic; it is secreted from the stomach and

GPR54 and Kisspeptins,GPR54 have hypogonadotropic hypogonadism characterized byabsence of sexual maturation and low levels of gonadotropic hormones (LH and FSH). The ligand for GPR54is encoded by the . gene, which produces a 54-amino-acid peptide(metastin or kisspeptin-54) that can be cleaved into shorter peptides (kissp

The Melanin-Concentrating Hormone System and Its Physiological Functions,so abundantly present in mammalian neurons andexpressed in the lateral hypothalamus and zona incerta, brain regions that are known to be at the centerof feeding behavior. MCH binds to and activates two G protein-coupled receptors, MCH1R and MCH2R. AlthoughMCH2R is non-functional in rodents, genetic

Biological Function of Prokineticins,ly homologous G protein-coupled receptors. Prokineticinspossess a unique structural motif of five disulfide bonds and conserved N-terminal stretches. Diversebiological functions, ranging from development to adult physiology, have been attributed to prokineticins.Herein we provide an overview of curr

Relaxin-3, INSL5, and Their Receptors, characteristic disulfide links connecting A andB peptide chains. R3 has nearly exclusive expression in the brainstem. It was demonstrated to be an additionalligand for the relaxin receptor LGR7, which is a class-C hormone receptor type G-protein coupled receptor(GPCR). We recently identified R3 as
