描述Advances in biochemistry, cell biology, genome-wide mutagenesis- coupled with molecular technology, including gene microarrayand transgenic and knock-out animals - have been instrumentalin understanding the cellular processes and molecular pathways ofself-tolerance and autoimmune diseases. The molecular definition ofthese pathways and processes has led to novel treatments for certainauto-immune diseases that are based on the pathogenesis of diseasesrather than on broad-spectrum immunosuppression. This book reviewsmany of these current developments and proposes future novelapproaches for understanding the pathogenesis of auto-immune diseasesand designing novel therapy. This book covers three major areas ofauto-immunity: the basic mechanisms of immunological tolerance, pathogenesis of auto-immune diseases, and some novel therapies. .This book should be useful for immunologists, molecular biologists, rheumatologists, and clinical scientists.

Cytokine Blockade in Rheumatoid Arthritis review the rationale for anti TNFa therapy, the current status of therapy and focus on the regulation of TNFa production in rheumatoid synovium. New approaches to studying TNF regulation in RA and of elucidating the controversial role of T cells in this complex disease will be described.

Peripheral Tolerance and Organ Specific Autoimmunityd receptor-deletion whereby immature B but not T cells change their receptors when confronted with antigen, perhaps because the antigen keeps the rearrangement process ongoing whereby already productive genes are deleted and new ones are being generated. Thus we have clonal or cellular deletion as w
