描述In 1980. a distinguished group of scientists gathered In Washington. D. C. for an International Symposium on Aging and Cancer. Among the recommendations of this Symposium was to convene a future meeting to discuss the molecular basis for Interrelationships between aging and cancer when the appropriate scientific knowledge was available. That same year. the 13th Jerusalem Symposium on Quantum Chemistry and Biochemistry entitled ·Carcl nogenesls : Fundamental Mechanisms and Environmental Effects·. was held. attended by some 50 International authorities In this field. At this meeting. It became clear that the fundamental process of carcinogenesis 15 Intimately associated with differentiation. which must also be mechanistically related to aging. It was therefore proposed that the next Jerusalem Symposium on Cancer could provide the appropriate forum for the study on the Interrelationship among cancer. aging and differentiation. The Impressive advances In our knowledge of the nature of the genome through molecular genetic and physical chemical techniques have now provided the opportunity to examine the Interrelationships between these complex biolo gical processes. Transforming Genes of Human Malignancies, tumors has also revealed that the movement of a specific cellular gene, again a homologue of a retroviral transforming gene, to a new regulatory environment alters its expression, thus contributing to tumor development.

Intermediates in Transcription Initiation and Propagation, essed copy of that gene renders the cell line less oncogenetic, probably by virtue of the body’s ability to mount an immunological defense. Altered gene expression seems likely to be important in the aging process as well, although genes specifically related to normal aging seem not yet to have been found.

Regulators of Growth, Differentiation and the Reversal of Malignancy, genes) there are other genes (soncogenes) that can suppress the action of oncogenes, that reversion does not have to restore all the normal controls, and that the stopping of cell multiplication by inducing differentiation to mature cells can by-pass the genetic abnormalities that give rise to malignancy.

Macromolecular Correlates of Cellular Senescence and Cancer,The consequence of both processes is increasing heterogeneity of the cell population, in its DNA structure and potential for gene expression. Such heterogeneity may underlie the age-dependent incidence of clonal-origin diseases such as cancer.

Deceleration of Cancer Mortality Rates with Age and Time, between cancer and aging, or a simple association between cancer and cumulative exogenous or endogenous factors. Cancer deaths can be expected to represent an ever decreasing proportion of total mortality although a slight rise in absolute numbers should persist.

Clinical Aspects of Cancer in the Elderly, ness. Patient management must include assessement of general functional and physiologic status and expected survival rates. Surgery, radiation therapy and chemotherapy can be utilized effectively, but individualization of treatment may be necessary. Psychosocial support and measures to maximize quality as well as quantity of life are important.
