Fungal Transposable Elementsause mutations makes them an interesting tool. The use of TEs for random mutagenesis would aid gene characterization in many filamentous fungi for instance in pathogenic or biotechnological important strains.

In Vivo Targeted Mutagenesis in Yeast Using TaGTEAMone repair via homologous recombination. Here we describe detailed protocols for implementing TaGTEAM in yeast, suitability of the technique for various evolutionary engineering goals, and considerations for porting TaGTEAM to other fungal species and beyond.

Transformation of Ascomycetous Fungi Using Autonomously Replicating Vectorsefold compared with a genome-integrating vector. The AMA1 vectors were maintained extrachromosomally in most transformants. In addition, co-transformation using multiple AMA1-bearing vectors was also successful. AMA1-bearing vectors are a useful addition to the experimental toolbox for many transformation experiments.

Deformable Model-Based Image Registration, for recessive RIP-deficient mutants; demonstrate meiotic silencing by unpaired DNA; and construct novel mutant strains. We can now make . strains in which transforming DNA integrates only by homologous recombination and use them to create knock-out mutants, possibly with DNA amplicons developed for making knock-outs in ..

Anastomosis and Heterokaryon Formationycelia in biotechnological applications. Recent years have seen a great revival of interest in the process of hyphal fusion, cummulating in the conclusion that we are currently still far from fully understanding the biology or appreciating the rich applicable potential of this fascinating process.
