NPY-mRFP Rat Basophilic Leukemia (RBL) Reporter: A Novel, Fast Reporter of Basophil/Mast Cell Degranization of the fluorescent protein in granules.  As NPY-mRFP is preformed in granules, the reporter system activation can be assessed using fluorescence measurements after as soon as 45–60 min, as described in this chapter, without the need to add any substrates.

With the method presented here, eight million mast cells can be cultured from ca. one million stem cells purified from one unit (450 mL) of human peripheral blood. Culture with IgE and IL4 optimizes an allergic phenotype of the mast cells.

The Mandel’shtam-Papalexi School: ,l functions have allowed better understanding of the function of mast cells and mast cell-mediated disorders. In this unit, we describe a protocol used for the generation and culture of peripheral CD34. stem cell-derived mast cells (PSCMCs). It provides a useful tool for the investigation of the biology of human MCs in vitro.
