Analysis of Phenylthiohydantoin Amino Acids by HPLC. It is the only technique commonly used for PTH analysis that can resolve all of the PTH amino acids in a single chromatographic run. Because it provides quantitative data at the pmol level, HPLC is also the only available method suitable for microsequencing that uses the new generation of automated Edman sequencers (.,.).

Manual Edman Sequencing Systementific discipline, existing as it does merely to supply probes to the molecular geneticists and occasionally to check their DNA reading frames, and one has a method in deep psychological crisis. Fortunately, manual sequencing can provide any of the structural data that requires or advantageously utilizes protein chemistry.

COOH-Terminal Analysis of Polypeptides by Chemical and Chromatographic Methodsrminal modifications, especially for many regulatory pep-tides (.,.). Thus, whenever Edman degradation is used as the prinicpal method of complete amino acid sequence determination, it must be supplemented by COOH-terminal analysis.
