Following the work of Brefeld, Lister and Koch, it became possible to study organisms in pure culture, but although this greatly simplified the task of observing and classifying organisms, it meant that they were studied under conditions that were vastly different from those that obtain in natural environments.

Moreover, the osmotic barrier can be broken by treating micro-organisms with lipid solvents, such as aqueous butanol, which cause the release of low molecular weight compounds from organisms. Microscopical observations on plasmolysable micro-organisms also indicate that the osmotic barrier resides in the cytoplasmic membrane.

Kai Wei,Qing Li,Yi Yao,Yeqing Sunntities of compounds that could be used in synthesis of new cell material, which indicates that there is extremely little wastage during cell metabolism. There are many other examples that could be quoted to show the remarkable degree of control and economy that operates in the metabolism of micro-organisms.
